Leucovorin as a Treatment Option for Autism

Leucovorin, also known as folinic acid, is a special form of folate (Vitamin B9). Unlike regular folic acid, it is already in an active form that the body and brain can use right away.

What is Leucovorin?

Leucovorin, also known as folinic acid, is a special form of folate (Vitamin B9). Unlike regular folic acid, it is already in an active form that the body and brain can use right away.

Why is it Considered for Autism?

Some children with autism spectrum disorder (ASD) may have difficulty getting folate into their brains. This can happen because of folate receptor antibodies (FRα) that block folate from crossing into the central nervous system.

This condition is called Cerebral Folate Deficiency (CFD).
Leucovorin can bypass this block and help restore healthy folate levels in the brain.

What Does the Research Say?

Studies have shown that high-dose leucovorin may:

  • Improve language and communication skills
  • Support better social interaction
  • Reduce irritability and hyperactivity in some children

In particular, children who test positive for FRα antibodies seem to benefit the most.

How is it Given?

  • Usually prescribed in higher doses than standard supplements
  • Often given as tablets or liquid suspension
  • Dosing and monitoring must always be done by a qualified healthcare provider

This is where a compounding pharmacy plays a role—formulating leucovorin into child-friendly liquid suspensions or flavored doses to make it easier for kids to take. Our compounded formulation for Leucovorin is Lactose free and dye free.  Our Leucovorin USP has been tested for potency and purity at the highest standard.  When our patients’ detoxification system is compromised, we need to use the cleanest product possible.  This is our promise to you!

Is it Safe?

Leucovorin is generally well tolerated. Some children may experience:

  • Mild hyperactivity or agitation
  • Stomach upset (nausea, constipation, or diarrhea)
  • Rare allergic reactions

Most side effects are manageable and often temporary

Key Takeaways for Parents

  • Leucovorin is not a cure for autism, but it may support improvements in certain symptoms.
  • Works best for children with cerebral folate deficiency or FRα antibodies.
  • Talk to your child’s doctor about whether leucovorin might be appropriate.
  • Compounding pharmacies can help by preparing leucovorin in personalized, child-friendly formulations.
